Comparative genomic hybridization analysis of invasive ductal breast carcinomas in the Chinese population

چکیده
Breast cancer is the most common malignancy in Chinese women. The aim of the present study was to investigate the genetic alterations that occur in breast cancer cells in Chinese women. Comparative genomic hybridization (CGH) analysis was performed on 34 tumors obtained from patients with primary invasive ductal breast carcinoma (IDC). Recurrent genetic alterations in breast cancer include gains on chromosomes 1q (59%), 16p (50%), 17q (44%), 8q (38%), 11q (32%), 20q (32%), 1p (24%), 20p (24%), 19q (21%) and 19p (18%). Losses are common on chromosomes 6q (15%), 8p (12%), 18 (12%), 4q (9%), X (9%) and 17p (9%). In the present study, high-level amplifications were observed on chromosomes 1q32, 8p, 11q13, 17q and 20q. Overall, the chromosomal DNA gains observed were consistent with the changes reported in Caucasian populations. However, the incidence of chromosomal DNA loss was lower in the present study compared with the incidence reported in the literature. The present results demonstrate the pattern of chromosomal imbalances in the invasive ductal breast carcinomas of Chinese females.

A preliminary study of microsatellite instability analysis in different genotypes of p53 codon 72 in breast invasive ductal carcinomas
Abstract Background: The polymorphic variants at codon 72 of the p53 gene, encoding either proline or arginine at residue 72, produce marked change in the structure of p53. From the evidence that the DNAmismatch repair system and p53 interact to maintain genomic integrity, we hypothesized that the codon 72 variation may influence the prevalence of microsatellite instability a featur...
متن کاملGenomic imbalances detected by comparative genomic hybridization are prognostic markers in invasive ductal breast carcinomas.
AIMS The aim of this work is the study of the prognostic significance of the chromosomal aberrations described in a series of invasive ductal breast carcinomas. METHODS AND RESULTS We analysed by comparative genomic hybridization a group of 70 formalin-fixed paraffin-embedded invasive ductal breast carcinomas. Aberrations showed a frequency similar to previous studies using frozen tumours. In...
متن کاملبررسی انحرافهای کروموزومی در کارسینوم داکتال مهاجم پستان به روش هیبریدیزاسیون ژنومی مقایسهای
Background: Nonlethal genetic damage is the basis for carcinogenesis. As various gene aberrations accumulate, malignant tumors are formed, regardless of whether the genetic damage is subtle or large enough to be distinguished in a karyotype. The study of chromosomal changes in tumor cells is important in the identification of oncogenes and tumor suppressor genes by molecular cloning of genes in...
متن کاملEvaluation Of The Androgen Receptor Status In Invasive Ductal Carcinoma Of Breast
Background and Objective: Determination of hormone receptor status in the management of breast cancer is well-established. The aim of this study was to evaluate the frequency of androgen receptor (AR) expression in invasive ductal carcinoma of breast. Materials and Methods: For this purpose, 55 cases of invasive ductal breast carcinoma were examined using a monoclonal antibody against AR on...
متن کاملCombined cDNA array comparative genomic hybridization and serial analysis of gene expression analysis of breast tumor progression.
To identify genetic changes involved in the progression of breast carcinoma, we did cDNA array comparative genomic hybridization (CGH) on a panel of breast tumors, including 10 ductal carcinoma in situ (DCIS), 18 invasive breast carcinomas, and two lymph node metastases. We identified 49 minimal commonly amplified regions (MCRs) that included known (1q, 8q24, 11q13, 17q21-q23, and 20q13) and se...
